Obiettivi dell'attività di ricerca
- valutazione, tramite tecniche di proteomica associate ad anticorpi contro epitopi fosforilati di proteine di segnale, di 1) identità, ampiezza e localizzazione dei segnali intracellulari in cellule normali in risposta a stimoli fisiologici, 2) le loro alterazioni in condizioni patologiche 3) lefficacia di trattamenti farmacologici nel ripristinare il corretto segnale e quindi il processo biologico/fisiologico.
- dissezione dei principali meccanismi di trasduzione del segnale che regolano la sopravvivenza cellulare e studio dei circuiti di feedback che favoriscono la plasticità delle cellule tumorali e la farmacoresistenza, con particolare interesse rivolto alla via di segnalazione PI3K/mTOR, ai suoi effettori/substrati e ai regolatori PTEN/PHLPPs.
- identificazione di nodi metabolici a valle di vie di segnale oncogeniche, che favoriscono la progressione tumorale e che sono considerati promettenti target terapeutici.
- valutazione del ruolo della nicchia midollare ipossica nel mantenimento delle cellule staminali leucemiche in quella condizione di quiescenza che le rende meno sensibili alla terapia convenzionale.
